Via my experience with Sasha Gunters's src.rpms for RHEL  and even xen-3.4.3-X-fc13.src.rpm on F12 behave same way. I mean after install libvirt related stuff on Fedora 12, virt-manager appears to be affected exactly you wrote.

 You may double check it :-
1. Install clean CentOS 5.4
2. Enable www.gitco's  repo 3.4.2
3. yum groupinstall Virtualization

That's why it was suggested to run virt-install for creating HVM S10U8.
Virt-manager may be used for managing any kind of DomUs , but only virt-install
utility stays completely functional in regards of setups. Issue was fixed only on F13 (Libvirt 0.8.2)

BTW , zfs-24 support patch by Mark Johnson  may be back ported to 3.4.3 gitco's
src.rpm . Gitco's Xen 3.4.3 rpms rebuilt and installed having active repo 3.4.3
to run  OSOL 134 as PV Guest on CentOS xen-kernel.

After installing to Xen 3.4.2, I am seeing the "virt-manager" GUI not working properly.  Before re-installing, virt-manager would ask for root password before executing, to give itself proper privileges for creating new VMs and such; now it no longer does, and cannot create new VMs.  When running as root, it then allows me to create new VMs, but only using a network install (http,ftp,nfs), not using either local media or a network pxfe boot.

Is this normal, or did I do something wrong when installing to 3.4.2?

Setup Xen 3.4.2 & Libvirt 0.6.3 on CentOS 5.3 (2.6.18-194.11.1.el5xen)
1.

Then reboot with entry:-
